Sinergisme Metabolit Sekunder dalam Pangan Fungsional: Mekanisme dan Implikasi Kesehatan

Manfaat kesehatan dari pangan fungsional sebagian besar dikaitkan dengan kandungan metabolit sekundernya yang melimpah. Namun, semakin banyak bukti ilmiah menunjukkan bahwa efek biologis dari pangan tersebut tidak dapat dijelaskan sepenuhnya oleh aksi senyawa tunggal yang terisolasi. Sebaliknya, konsep sinergisme — yaitu ketika efek gabungan dari beberapa konstituen bioaktif lebih besar daripada jumlah efek individualnya — kini semakin diakui sebagai prinsip dasar yang penting. Tinjauan ini secara sistematis mensintesis pengetahuan terkini mengenai mekanisme yang mendasari sinergisme antar metabolit sekunder dalam matriks pangan fungsional dan menguraikan dua kategori mekanistik utama, yaitu: (1) Sinergisme farmakokinetik, yang melibatkan peningkatan bioavailabilitas melalui penghambatan enzim metabolik, modulasi transporter absorpsi, serta biotransformasi oleh mikrobiota usus; dan (2) Sinergisme farmakodinamik, yang mencakup interaksi multi-target dalam jalur patologis, efek antioksidan yang saling melengkapi, serta stabilisasi sistem seluler serta melakukan evaluasi kritis terhadap bukti empiris mengenai sinergisme tersebut pada kelompok pangan fungsional utama, termasuk rempah-rempah (misalnya kombinasi kurkumin–piperin), buah beri, teh hijau, dan tanaman allium. Akhirnya, kami membahas implikasi temuan ini terhadap perancangan penelitian di masa depan, pengembangan pangan fungsional berbasis bukti ilmiah, serta perumusan rekomendasi diet untuk pencegahan penyakit kronis. Mengadopsi perspektif sinergistik yang kompleks ini sangat penting untuk memajukan ilmu gizi dan membuka potensi penuh pangan sebagai agen terapeutik alami.
